Every Nanometer Counts: Optimizing Sample Preparation for STED Microscopy

Abstract:
Stimulated emission depletion (STED) microscopy enables fluorescence imaging at nanometer resolution by overcoming the diffraction limit of light. However, the same optical precision that allows sub-50nm resolution also imposes stringent demands on sample preparation. This article provides a comprehensive overview of the key considerations required for successful STED imaging, including fluorophore selection, labeling strategies, fixation, permeabilization, mounting media, and live-cell compatibility. We highlight how even minor deviations from optimized workflows, such as linkage error, refractive index mismatch, or autofluorescence, can compromise resolution and accuracy. Practical strategies are discussed for mitigating common pitfalls, improving signal fidelity, and adapting protocols to the unique requirements of STED. By emphasizing a preventive, resolution-aware approach to sample prep, this guide aims to support researchers in achieving reproducible, artifact-free, super-resolution STED imaging.
