Emerging Trends in Super-resolution Imaging: How Lasers Light the Way

Abstract:
Super-resolution imaging has transformed the way we see the nanoscale world, allowing us to defeat the diffraction limit of light and observe the behaviors of single molecules with unprecedented detail. Lasers are key to the evolution of this ever-changing field. By modulating polarization, wavelength, spatial excitation profiles and temporal excitation profiles, researchers have pushed super-resolution imaging into new frontiers in which we are no longer beholden to special classes of photoswitchable probes or limited to fluorescence imaging. This chapter will describe advances in super-resolution imaging enabled by lasers, focusing specifically on single molecule localization microscopy (SMLM) and stimulated emission depletion (STED) variants.
